缺血后处理对猪非梗死区心肌细胞凋亡的影响

摘要:
[摘要]目的 探讨缺血后处理对猪非梗死区心肌细胞凋亡的影响.方法 滇南小耳猪15只,随机分为3组.(1) 假手术组(S组,n=5),未行冠脉封堵及心肌缺血;(2)缺血再灌注组(I/R组,n=5),心肌缺血60 min后,不行缺血后处理,直接撤除球囊;(3) 缺血后处理组(Postcond组,n=5),心肌缺血60 min后,通过球囊充气和放气造成8次30 s/30 s短暂缺血再灌注.再灌注72 h采用2,3,5氯化三苯基氯四氮唑(TTC)染色确定心肌梗死面积;在猪非心肌梗死区,采用TUNEL法检测心肌凋亡细胞及免疫组化检测Bcl-2和Bax的蛋白表达.结果 再灌注72 h,缺血后处理组心肌梗塞面积、非心肌梗死区细胞凋亡指数及Bax的蛋白表达和Bax/Bcl-2的比值明显低于再灌注组(P<0.05).结论 缺血后处理可能通过减轻非梗死区心肌细胞凋亡发挥心肌保护作用.

Effects of Ischemic Postconditioning on Myocardial Apoptosis in Non-infarct Zone in Pigs with Myocardium

Abstract:
[Abstract]Objective To investigate the effect of ischemic postconditioning on myocardial apoptosis in non-infarct zone in pigs with myocardium.Methods Fifteen Diannan small-ear pigs were divided into three groups(n=5 pre group) randomly:(1)Sham-operated(S)group:A balloon catheter was positioned in the mid-distal left anterior descending(LAD)via right femoral artery under fluoroscopic guidance,without ischemia;(2)Ischemia-reperfusion (I/R) group: the balloon was inflated and occluded the LAD for 60 minutes;(3)ischemic postconditioning(Postcond)group: pigs were treated the same as I/R group and 8 cycles of 30 s R and 30 s I before reperfusion. After 72 h of reperfusion,infarction size were measured by 2,3,5-triphenyltetrazolium chloride(TTC)staining.The apoptotic cells in the non-infarct zone myocardium was determined with terminal deoxynucleotidyl transferase-mediated dUTP-fluorescence in nick end(TUNEL) method,and apoptosis-related molecules were studied by immunohistochemical analysis. Result After 72h of reperfusion,the infarction size,apoptotic index(AI)of non-infarcted zone,the levels of Bax protein expression,and Bax/Bcl-2 ratio were significantly lower in Postcond group than those in I/R group(P<0.05).Conclusion Ischemic postconditioning can reduce myocardial apoptosis in non-infarct zone and maybe play a myocardial protection effect.
