应用组织芯片技术对FHIT基因在90例生存期肝癌的表达

[摘要]目的 探讨应用组织芯片技术对FHIT基因在90例生存期肝癌手术标本中的表达差异.方法 收集2010年1月至2013年9月90例临床资料和随访记录完整的原发性肝癌患者的手术标本,以相应的癌旁组织为对照,以免疫组化染色制作组织芯片,观测上述标本中FHIT基因表达差异.结果 FHIT基因在本实验组的人肝癌手术标本中的低表达是高概率事件(67/90;74.4%),同时癌旁组织的FHIT基因表达显著高于肝癌细胞(P<0.05)且组织芯片免疫组化染色定量分析FHIT基因在本实验组的肝癌组织和癌旁组织中的表达差异有统计学意义(P<0.05).结论 FHIT基因是一种抑癌基因,其在肝癌细胞中的低表达可提示肿瘤恶性程度增加,免疫组化检测FHIT基因有助于肝癌的恶性程度判断和术后生存期预后的合理评估.

[Abstract]Objective To explore the expressions differences of the FHIT gene in 90 human hepatocellular carcinoma tissues with tissue chip method. Methods 90 cases of patients with primary liver cancer of complete the clinic pathological data and its surgical specimens to keep in the archives with paraffin blocks were collected. FHIT gene expressional differences in the above specimens were determined by IHC-tissue chip. Results Whether qualitative or quantitative analysis,IHC test results showed that there was a significant difference in expressions of FHIT protein between hepatocellular carcinoma tissue and para-carcinoma tissue. The positive expression of FHIT protein in para-carcinoma tissue was significantly higher than hepatocellular carcinoma tissue(P<0.05). Conclusions FHIT gene is a cancer suppressor gene and poor expression of FHIT gene is correlated with malignant differentiation of liver cancer. Immunohistochemical method is helpful to assist the early diagnosis and make a reasonable appraisal of their prognosis.
