NOXA、PUMA等基因在乳腺癌中的表达及其意义

摘要:
[摘要]目的 比较癌组织与正常组织中MDM2、PCNA、NOXA、PUMA表达的差异,了解乳腺癌与表达的关系. 方法 通过SYBR Green实时荧光定量PCR方法检测乳腺癌组织和正常乳腺组织中MDM2、PCNA、NOXA、PUMA 4个基因的含量.结果(1)NOXA、和PUMA在乳腺癌组织中表达升高(P<0.05);(2)PCNA和NOXA的表达存在正相关关系(r=0.82,P<0.05)); PCNA和MDM2的表达存在正相关关系(r = 0.80,P<0.05);PUMA和MDM2的表达存在正相关关系(r=0.82,P<0.05) . 结论 NOXA、PUMA在乳腺癌组织中处于高表达状态,4个基因表达与乳腺癌有关.

Abstract:
[Abstract]Objective To study the expression characteristics of PCNA, NOXA, PUMA and MDM2 in breast cancer. Methods The cancer tissue and adjacent normal tissue were collected from 35 patients with breast cancer. The total RNA was extracted using TRIzol and the expression levels of MDM2, PCNA, NOXA and PUMA were detected by real-time quantitative PCR. Results NOXA and PUMA levels in cancer tissue were significantly higher than those in adjacent normal tissue(P < 0.05). There were significant positive correlations between the expressions of PCNA and NOXA (r=0.82, P < 0.01), PCNA and MDM2 (r=0.80, P < 0.01), and PUMA and MDM2(r=0.82,P < 0.01)in adjacent normal tissue. Conclusion The levels of NOXA and PUMA were high expression in cancer tissue, and the expressions of all 4 genes showed relations with breast cancer.
