Let-7在正常乳腺组织和乳腺癌组织中的表达

摘要:
[摘要]目的 研究Let-7基因在乳腺癌中的表达特征.方法 随机抽取28例乳腺癌患者收集其癌组织和癌旁正常组织,利用TRIzol提取总RNA,实时荧光定量PCR检测基因表达量.结果 Let-7前体在癌组织中的相对表达量(9.65±2.31),低于癌旁正常组织(10.0±52.81),P=0.048,而Let-7基因则无显著变化.前体Let-7与最长行经(日)之间有依存关系(b=0.875,P=0.020);初潮年龄与前体Let-7的表达在癌组织中的呈负相关(r=-0.484,P=0.049), 在正常组织中呈正相关(r=0.502,P=0.048).结论 Let-7前体在癌组织中的表达低于正常组织, 最长行经天数越长则Let-7表达越高.

[Abstract]Objective To study the expression characteristics of Let-7 genes in breast cancer. Methods Twenty-eight patients with breast cancer were randomly selected,and their cancer tissue and adjacent normal tissue were collected. TRIzol was used to extract the total RNA and real-time quantitative PCR was used to detect the relative gene expression levels. Results The expression of precursor Let-7 in cancer tissue was(9.65±2.31),which was lower than that in normal tissue(10.05±2.81 ),P = 0.048. Precursor Let-7 had dependence relationship with the long menstruation(b = 0.816,P = 0.029). The menarche age showed a positive correlation with precursor Let-7 in normal tissue(r = 0.502,P = 0.048)and a negative correlation in cancer tissue(r = -0.484,P = 0.049). Conclusions The expression of precursor Let-7 in cancer tissue is lower than that in normal tissue. The period of menstruation is a protective factor to breast cancer.
